Nöbet'in 1. Bölümünde; Jandarma Komutanı Yüzbaşı Orhan Korkut, teröristlerle yaşanan sıcak çatışmada askerini korumak için kendi canını feda ederek, şehit düşer. Onun yerine gelen Binbaşı Barış, evlatlarının başını öne eğmemeye ve Orhan Yüzbaşı'yı şehit eden hainleri bulmaya yemin etmiştir! Jandarma Özel Harekât timinin komutanı Jandarma Yüzbaşı Yiğit Alkan ise dağlarda Orhan Yüzbaşı'nın şehit olduğu saldırının da arkasında olan Jiyan kod adlı teröristin peşindedir. Devre arkadaşı ve yakın dostunu öldüren teröristi bulmaya o da yeminlidir. Bu iki gözüpek asker, Hatay'da huzursuzluk çıkarmak isteyenlere karşı birlikte mücadele edeceklerdir. Barış Binbaşı daha bölgeye adım atar atmaz, bölgenin zenginlerinden Ziya Cevran huzursuzluk çıkarmıştır. Ziya Cevran ve oğlu Ferit'in daha birçok sorun çıkaracağı ve kontrol altında tutulmaları gerektiği aşikârdır. Tüm bunlar olurken Yiğit Yüzbaşı, Jiyan ile ilgili bir istihbarat alır, timiyle beraber peşine düşer. Barış Binbaşı da aynı terör örgütünün ahin bir plan peşinde olduğunu fark eder ve bölgenin en ıssız vadisinde Jandarma Kuvvetleri'ne kurulan pusuyla karşı karşıya kalır…
